"Cyclophosphamide is a chemotherapy drug widely used to treat various types of cancer. However, its use can cause hemorrhagic cystitis of the bladder as a side effect. Petai cina seeds contain sulfhydryl compounds that have the potential to be used as an alternative to prevent hemorrhagic cystitis by inducing glutathione production and metabolism," said Dr. apt. Sri Wardatun, M.Farm., at the Doctoral Promotion event for Pharmaceutical Sciences at the Faculty of Pharmacy, University of Indonesia (FFUI), Wednesday (July 27, 2022).

The Open Doctoral Promotion Session was opened by Prof. Dr. apt. Arry Yanuar, M.Si. as the Chair of the Session. Next, in the material presentation session, Sri began the discussion on the objectives and analysis methods in her research entitled "Analysis of 4-Hydroxycyclophosphamide and 3-Hydroxypropyl Mercapturic Acid after Administration of Chinese Petai Seed Extract to Cyclophosphamide-Induced Hemorrhagic Cystitis Rats".
In her research, Sri analyzed 3-hydroxypropyl mercapturic acid in urine and 4-hydroxycyclophosphamide in plasma to assess the potential of Chinese Petai Seed extract in preventing hemorrhagic cystitis in cyclophosphamide-induced mice. "To achieve the objectives of the analysis, extraction solvent optimization was carried out to obtain the maximum solvent concentration that can produce maximum sulfhydryl compounds and minimum mimosine. Next, standardization of the extract, validation of the analysis method using KCKUT SM/SM, and analysis of compound levels in vivo were carried out to obtain the activity of Chinese Petai Seed extract," she explained.
The results of this study indicate that 70% ethanol is the solvent that produces extracts with the highest sulfhydryl content and the lowest mimosine content in petai cina seeds that have been soaked for 24 hours. The 70% ethanol extract of petai cina seeds meets the quality standards for traditional medicinal extracts. The analysis method for 3-HPMA in urine and 4-hydroxycyclophosphamide in plasma using KCKUT SM/SM has been validated according to the FDA, 2018 and EMA, 2011 references and can be applied in vivo analysis. Administration of 70% ethanol extract of petai cina seeds can increase 3-HPMA levels in urine and does not affect the levels of the active metabolite 4-hydroxycyclophosphamide in plasma.

"Administering petai cina seed extract had a positive effect on bladder histopathology results and the hematological profile of mice. Petai cina seed extract has the potential to be used as an alternative to prevent hemorrhagic cystitis in patients with cyclophosphamide," said Sri.
Based on the submitted dissertation and the examiners' consideration of Sri's arguments, the examining team decided to elevate Apt. Sri Wardatun, M.Farm., to a Doctorate in Pharmaceutical Sciences with cum laude graduation.
Prof. Dr. apt. Yahdiana Harahap, MS, as the Promoter, is giving a speech (Source: UI Faculty of Pharmacy Public Relations Documentation)
"Mrs. Sri's research is extraordinarily complex and she was able to complete it well. This research was chosen because cyclophosphamide, which is a chemotherapy drug, still has many side effects, especially hemorrhagic cystitis in the bladder. The administration of Chinese Petai Seed extract has a positive effect on the results of histopathology and hematology tests, so it has great potential to be used as an alternative in preventing hemorrhagic cystitis in the use of cyclophosphamide. Mrs. Sri has also developed a method of two biological matrices in plasma and urine which was then validated according to international guidelines , and that is extraordinary," said Prof. Yahdiana in his speech as Promoter. In his speech, Prof. Yahdiana also hoped that the research conducted by Dr. Sri can be continued to help clinicians in treating hemorrhagic cystitis.
